This project will explore the effect of CRISPR/Cas9 mediated mutations in the NXF1 gene on the outgrowth and function of primary hippocampal and cortical neurons in culture systems.
NXF genes are involved in trafficking of mRNA in RNAgranules and cooperate with FMRP, a gene that is mutated in the most commonform of male mental retardation. Mutation in NXF genes have also been linked tomental retardation.
The project will use neuronal imaging techniqesas well as RNA Seq, using both Illumina and Oxford Nanopore (Minion)technologies to further explore the hypothesis that NXF1 is important forneuronal function and synaptic plasticity.
We hope that this will establish a fruitful collaboration between RNA biologists and Neurobiologists that will generate the data necessary for a major NIH application and for publication in a high quality journal.
